Preventive Effect of Natural Herb Extracts With PPAR ¥á/¥ã Dual Agonistic Effect on Photo-Aging

Abstract
We found that extracts of natural herbs such as Kochia Scoparia and Rosa multiflora have PPAR ¥á/¥ã dual agonistic effect. Therefore, we investigate whether they have anti-aging effect on photo-aging skin. Hairless mice were irradiated with ultraviolet (UV) A 14J/cm2 and UVB 40mJ/cm2 three times a week for 8 weeks. Natural herb extracts with PPAR ¥á/¥ã dual agonistic effect were topically applied on dorsal skin of photo-aging mice twice a day for 8 weeks. Tesaglitazar, a known PPAR ¥á/¥ã agonist, were applied as a positive control, and vehicle (PG:EtOH=7:3, v/v) as a negative control. Stratum corneum (SC) integrity and barrier recovery rate as well as anti-aging dermal effects including dermal thickness, dermal collagen density, mRNA and protein expression of collagen and collagenase were measured respectively and compared. The natural herb extracts with PPAR ¥á/¥ã dual agonistic effect enhanced skin barrier function by increasing SC integrity and barrier recovery rate. In addition, they increased dermal thickness and dermal collagen density without the change of matrix metalloproteinase 13. Extracts of natural herbs such as Kochia Scoparia and Rosa multiflora with PPAR ¥á/¥ã dual agonistic effect could prevent the process of photo-aging by both of increasing collagen density due to collagen production and improving skin barrier function.
